This is a rich media that is used for recovery of transformed bacteria.

  • 20g tryptone
  • 5g yeast extract
  • 0.5g NaCl

Add the above to 950ml of H2O. Add 10ml of a 250mM KCl solution. pH the media to 7.0 with NaOH, then adjust the volume to 1 liter. Autoclave on cycle 3 (Keck Hall 2nd and 3rd floor autoclaves). After autoclaving, add 5ml/L of a sterile 2M MgCl2 solution. At this point, the media is SOB. To make SOC, add 20ml/L of sterile 1M glucose (or 18ml/L of sterile 20% glucose) to the media.
